Carbon dioxide (CO<sub>2<\/sub>) insufflation during laparoscopy may have an anti-inflammatory effect. We aimed to evaluate the effects of CO<sub>2<\/sub>-insufflation on experimental colitis.<\/p>\r\n<\/section>\r\n<section id=\"jpn312048-sec-0020\" class=\"article-section__content\">\r\n<h3 id=\"jpn312048-sec-0020-title\" class=\"article-section__sub-title section1\">Methods<\/h3>\r\n<p style=\"text-align: justify;\">Acute colitis was induced in 6-week-old Balb\/c mice by the administration of 2%-dextran sulfate-sodium (DSS) during 7 days (<i>n<\/i>\u2009=\u200945). On Day 4, two groups received intraperitoneal insufflation (duration: 30\u2009mn, pressure: 5 mmHg) of CO<sub>2<\/sub>\u00a0(\u201cDSS+CO<sub>2<\/sub>\u201d) or air (\u201cDSS+air\u201d). A group received no insufflation (\u201cDSS\u201d). Groups were compared for clinical severity using the disease activity index (DAI\u2014body weight loss, stool consistency, and bleeding), histological severity (histopathological activity index, colon length, and ulcerations), colonic mucosecretion, and inflammation.<\/p>\r\n<\/section>\r\n<section id=\"jpn312048-sec-0030\" class=\"article-section__content\">\r\n<h3 id=\"jpn312048-sec-0030-title\" class=\"article-section__sub-title section1\" style=\"text-align: justify;\">Results<\/h3>\r\n<p style=\"text-align: justify;\">DAI was significantly decreased in DSS+CO<sub>2<\/sub>\u00a0group, compared to DSS (<i>p<\/i>\u2009&lt;\u20090.0001) or DSS+air (<i>p<\/i>\u2009&lt;\u20090.0001) groups. Colon length was increased in DSS+CO<sub>2<\/sub>\u00a0treated mice compared to DSS (<i>p<\/i>\u2009=\u20090.0002). The histopathological activity index was lower in DSS+CO<sub>2<\/sub>\u00a0(vs. DSS,\u00a0<i>p<\/i>\u2009=\u20090.0059\/vs. DSS+air,\u00a0<i>p<\/i>\u2009=\u20090.0389), with decreased ulcerations (3.77 vs. 10.7, <i>p<\/i>\u2009=\u20090.0306), and persistent mucosecretion with increased mucin-secreting cells.<\/p>\r\n<\/section>\r\n<section id=\"jpn312048-sec-0040\" class=\"article-section__content\">\r\n<h3 id=\"jpn312048-sec-0040-title\" class=\"article-section__sub-title section1\" style=\"text-align: justify;\">Conclusions<\/h3>\r\n<p style=\"text-align: justify;\">CO<sub>2<\/sub>-insufflation attenuates DSS-induced colitis and improves both clinical and histological scores.
