Les voies de signalisation r\u00e9gulant l\u2019activation de cette oxydase dans les neutrophiles ont \u00e9t\u00e9 largement \u00e9tudi\u00e9es \u00e0 l\u2019aide d\u2019agonistes solubles, mais elles sont moins bien comprises lors de la phagocytose, une fonction fondamentale des neutrophiles.<\/p>\r\n<p style=\"text-align: justify;\" data-start=\"477\" data-end=\"940\">L\u2019objectif de cette \u00e9tude \u00e9tait d\u2019examiner la phosphorylation de la prot\u00e9ine cytosolique p47phox de l\u2019oxydase NADPH dans des neutrophiles humains stimul\u00e9s par du zymosan opsonis\u00e9 par du s\u00e9rum (OZ), qui induit la phagocytose, en utilisant des anticorps dirig\u00e9s contre des sites phosphoryl\u00e9s. Les r\u00e9sultats montrent que l\u2019OZ induit une phosphorylation rapide de p47phox aux r\u00e9sidus Ser304, Ser315, Ser320 et Ser328, suivie d\u2019une d\u00e9phosphorylation tout aussi rapide.<\/p>\r\n<p style=\"text-align: justify;\" data-start=\"942\" data-end=\"1304\">Fait int\u00e9ressant, malgr\u00e9 la nature transitoire de cette phosphorylation, l\u2019activit\u00e9 de l\u2019oxydase NADPH induite par l\u2019OZ est rest\u00e9e soutenue plus longtemps, aussi bien dans les cellules intactes que dans les membranes isol\u00e9es. La phosphorylation de p47phox induite par l\u2019OZ \u00e0 ces sites \u00e9tait d\u00e9pendante de la concentration et pr\u00e9c\u00e9dait l\u2019ingestion des particules.<\/p>\r\n<p style=\"text-align: justify;\" data-start=\"1306\" data-end=\"1553\">Le zymosan opsonis\u00e9 par des IgG ou du C3bi induisait \u00e9galement une phosphorylation et une d\u00e9phosphorylation rapides de p47phox aux r\u00e9sidus Ser304, Ser315, Ser320 et Ser328, ce qui sugg\u00e8re l\u2019implication des r\u00e9cepteurs Fc\u03b3R et CR3 dans ce processus.<\/p>\r\n<p style=\"text-align: justify;\" data-start=\"1555\" data-end=\"1743\">Des inhibiteurs des tyrosine kinases Src et Syk, de la PI3K, de la PLC, de la PLD, du calcium (Ca++) et de la PKC\u03b22 ont inhib\u00e9 la phosphorylation de p47phox induite par l\u2019OZ \u00e0 ces r\u00e9sidus.<\/p>\r\n<p style=\"text-align: justify;\" data-start=\"1745\" data-end=\"1776\">Ces r\u00e9sultats sugg\u00e8rent que :<\/p>\r\n\r\n<ol data-start=\"1777\" data-end=\"2211\" data-is-last-node=\"\" data-is-only-node=\"\">\r\n \t<li style=\"text-align: justify;\" data-start=\"1777\" data-end=\"1986\">\r\n<p data-start=\"1780\" data-end=\"1986\">la phosphorylation de p47phox aux r\u00e9sidus Ser304, Ser315, Ser320 et Ser328 induite par l\u2019OZ est n\u00e9cessaire pour initier l\u2019activation de l\u2019oxydase NADPH, mais pas pour son maintien pendant la phagocytose ;<\/p>\r\n<\/li>\r\n \t<li style=\"text-align: justify;\" data-start=\"1987\" data-end=\"2075\">\r\n<p data-start=\"1990\" data-end=\"2075\">les r\u00e9cepteurs membranaires Fc\u03b3R et CR3 sont impliqu\u00e9s dans cette phosphorylation ;<\/p>\r\n<\/li>\r\n \t<li data-start=\"2076\" data-end=\"2211\" data-is-last-node=\"\">\r\n<p style=\"text-align: justify;\" data-start=\"2079\" data-end=\"2211\" data-is-last-node=\"\">les kinases Src et Syk, la PI3K, la PLD, le calcium et la PKC\u03b22 contr\u00f4lent la phosphorylation de p47phox au cours de la phagocytose.<\/p>\r\n<\/li>\r\n<\/ol>\r\n<img class=\" wp-image-74682 aligncenter\" src=\"https:\/\/cri1149.fr\/wp-content\/uploads\/2025\/07\/blood-el-benna-2025-300x241.png\" alt=\"\" width=\"675\" height=\"542\" \/>","paragraphe_en":"","documents":null},"yoast_head":"<!-- This site is optimized with the Yoast SEO plugin v26.8 -
